# Single Cell Sample Prep Re-engineered

Ride the acoustic wave to enzyme-free tissue
dissociation with SimpleFlow TM

![Simpleflow utilizes bulk lateral ultrasound (BLU)](https://cellsonics.com/hs-fs/hubfs/Cellsonics%20-%202024/Images/Home/Simpleflow%20utilizes%20bulk%20lateral%20ultrasound%20(BLU).png?width=614&height=710&name=Simpleflow%20utilizes%20bulk%20lateral%20ultrasound%20(BLU).png)

In contrast to enzymes that digest biological material to release cells, **SimpleFlow** utilizes acoustic waves that dissociate more cells without bias.

**SimpleFlow** avoids the enzymes and heat typically used in traditional tissue dissociation methods, which are known to negatively impact the underlying biology and can selectively eliminate more sensitive cell types.

As a result, **SimpleFlow** sources cell populations that are more representative of the native tissue to ensure higher biological relevance.

**Dissociate tissue** without the use of enzymes

Obtain **high-yield and high-viability** single-cell
suspensions from a broad range of tissue types

Retain **native cell populations** present in tissue prior
to dissociation

Process a sample **in minutes** and simplify
cleanup procedures

Simply add tissue, mince, and tilt to transfer,
SimpleFlow's acoustics do the rest of the work.

Figure 1. A single cell RNA-seq comparison of whole mouse brain dissociated with SimpleFlow and a commercially available enzyme kit.

Tissue was dissociated on the SimpleFlow using the mouse brain protocol which ran for 4 minutes. The enzyme dissociation was performed according to the manufacturer's protocol.

Mouse brain dissociated with SimpleFlow demonstrated higher preservation of astrocytes and neurons. The enzymatic dissociated brain over-represents microglia, immune cells in the CNS that patrol the brain for damage and pathogens. This is indicative of stress and damage due to the dissociation process.

These findings are consistent with published scientific literature!

Left. A comparison of 100 mg of adult mouse brain dissociated by either SimpleFlow or a commercially available enzyme kit.  The count of live cells per mg of tissue was determined by volumetric flow cytometry. Live cells were identified as Propidium Iodide (PI) negative excluding debris.

Right. A comparison of 100 mg of adult mouse brain dissociated by either SimpleFlow or a commercially available enzyme kit.  The percent viability was determined by flow cytometry and calculated as Propidium Iodide (PI) negative cells divided by total cells excluding debris.

## Improve downstream single-cell analysis with well treated cells

##### Tissue dissociation, a crucial step in preparing single cell suspensions for Flow Cytometry or Single-cell RNA-seq (scRNA-seq), plays a pivotal role in single cell analysis.

##### Conventionally, enzymatic incubations at 37°C have been the go-to method for tissue dissociation, despite numerous scientific publications highlighting the significant adverse effects of heat and enzymes on the cellular biology of individual cells.

##### Enzymatic dissociation has been repeatedly demonstrated to introduce artificial alterations in gene expression patterns, negatively impacting cell surface markers and impeding the accurate selection of cell subtypes via flow cytometry and single cell RNA-seq.
