Tissue dissociation, a crucial step in preparing single cell suspensions for Flow Cytometry or Single-cell RNA-seq (scRNA-seq), plays a pivotal role in single cell analysis. Conventionally, enzymatic incubations at 37°C have been the go-to method for tissue dissociation, despite numerous scientific publications highlighting the significant adverse effects of heat and enzymes on the cellular biology of individual cells. Enzymatic dissociation has been repeatedly demonstrated to introduce artificial alterations in gene expression patterns, negatively impacting cell surface markers and impeding the accurate selection of cell subtypes via flow cytometry.

As an alternative to prolonged enzyme-based incubations at 37°C, the Cellsonics SimpleFlow introduces a rapid, enzyme-free tissue dissociation approach. SimpleFlow harnesses the power of Bulk Lateral Ultrasonic™ (BLU) energy, an acoustic energy-based technique that delicately pulses minced tissue to efficiently yield a single cell suspension. This dissociation process is brief, often completing within 2 minutes, all while maintaining the sample temperature at approximately 6°C. Data demonstrates that employing BLU technology creates a more favorable environment for the single cells destined for downstream applications such as scRNA-seq or precise cell selection via flow cytometry.
